Tadalafil is a carboline-based compound with vasodilator activity. It selectively inhibits cyclic guanosine monophosphate (cGMP), a precursor of specific phosphodiesterase type 5 (PDE-5). It is located in the smooth muscle of the cavernous tissue. Taking 시알리스 leads to prolonged muscle relaxation, vasodilation, and blood flow to this tissue. With its help, an erection of the penis is achieved. It is available in pill form and is a powerful sexual stimulant. It is available in pill form for oral administration. The distinctive pharmacological feature of Tadalafil is its longer half-life (17.5 hours) compared to Viagra and Levitra (4-5 hours). This longer period results in a longer duration of action and is partly responsible for the nickname 시알리스 - the "weekend pill".
How it works
The exact mechanism has not been established. It has been shown to result in decreased nerve activity and increased smooth muscle relaxation and tissue perfusion of the prostate and bladder. It does not directly cause a penile erection but affects the response caused by sexual stimulation. The physiological mechanism of penile erection involves the release of nitric oxide (NO) into the cavernous tissue during sexual stimulation. NO then activates the enzyme guanylate cyclase, leading to increased levels of cyclic guanosine monophosphate (cGMP), producing smooth muscle relaxation and blood flow to the cavernous tissue. Tadalafil enhances the effect of nitric oxide by inhibiting phosphodiesterase type 5 (PDE-5), which is responsible for the breakdown of cGMP. When sexual stimulation causes local release of NO, inhibition of PDE-5 by tadalafil causes increased levels of cGMP, which results in smooth muscle relaxation and blood flow to the penis. The increased blood flow causes a stronger erection for a long period of time. At recommended doses, it has no effect in the absence of sexual stimulation.
